A New Maraglas, D.e.r.® 732, Embedment for Electron Microscopy
نویسنده
چکیده
A new epoxy embedding mixture for biological material recently has been introduced by Freeman and Spurlock (1). This mixture consists of Mara-glas 655, Cardolite NC-513, dibutyl phthalate, and the curing agent benzyldimethylamine (BDMA). Maraglas has certain advantages over other resins and epoxies, notably, wide range of miscibility, low viscosity, ease of sectioning, good staining qualities, beam stability, and little background granularity. Although improvements have been made in the original technique (2), certain problems remain. The Maraglas mixture penetrates tissue very slowly, makes the tissue quite brittle, and has variable polymerization properties. In this paper the use and advantages of a new polyglycol type flexibilizer as a replacement for Cardolite NC-513 in a Maraglas 655 epoxy formulation will be presented.

A New Epoxy Embedment for Electron Microscopy
A new epoxy embedding mixture has been developed utilizing Maraglas 655 and Cardolite NC-513 with benzyldimethylamine (BDMA) as a curing agent. This epoxy mixture permits cellular preservation comparable to that obtained with Epon 812, ease of preparation of tissues, a wide range of miscibility, low viscosity, and, most important, ease of sectioning on a Porter-Blum microtome. In contrast to Ep...
متن کاملAn experimental study of axial load transfer mechanisms of cable bolts using axially split embedment apparatus
The load transfer mechanisms of cable bolts differ from those for normal rebar bolts. The cable bolts used in mines are basically steel strands with different constructions depending on the number of wires or elements and the way they are laid. Tendon bolts (rebar and cable) are normally evaluated for their strength and load transfer properties. The tendon strength can be evaluated by the tensi...
متن کاملMechanistic Features of Nanodiamonds in the Lapping of Magnetic Heads
Nanodiamonds, which are the main components of slurry in the precision lapping process of magnetic heads, play an important role in surface quality. This paper studies the mechanistic features of nanodiamond embedment into a Sn plate in the lapping process. This is the first study to develop mathematical models for nanodiamond embedment. Such models can predict the optimum parameters for partic...
متن کاملImaging cytoskeleton--mitochondrial membrane attachments by embedment-free electron microscopy of saponin-extracted cells.
Embedment-free electron microscopy images the cytoskeleton and nuclear matrix, which are very difficult to visualize in conventional electron micrographs. However, to be effective, cell structures must be depleted of soluble proteins, which otherwise shroud cell architecture. Nonionic detergents effect this extraction, releasing soluble proteins but also destroying all membranes. Saponin can pe...
متن کاملTechnical Modifications in Maraglas Embedding
Marglas 655 is proving to be a versatile embedding medium for biological electron microscopy (1-5) in that it provides favorable cutting qualities and excellent cellular preservation (Figs. 1, 2, 4). Since the publication of the original technic (1), we have found several useful technical modifications and developed methods for staining sections for light microscopy. Two technical difficulties ...
